I am currently on a one-year Single Entry Visa based on marriage. I'm wondering if it's possible for me to apply for a Multiple re-entry Permit at the Immigration Office in Chamchuri Square Building.

Chamchuri office wouldn't handle anything related to visas or extensions based on being married to a Thai. You have to go to the Chaengwattana office to get a re-entry permit on your Non-O-based visa or extension.
BTW, There is no such thing "one-year Single Entry Visa". Please check your visa and admitted-to-stay stamp and know what you really have.

- Multiple Entry Non-O visa based on being married to a Thai from the Thai embassy or consulate outside Thailand valid for 1 year with 90 days for each entry,
- Single Entry Non-O visa based on being married to a Thai from the Thai embassy or consulate outside Thailand valid for 3 months with 90 days for an entry,
for the prompt response. I got the "THAI WIFE" 1 year extension from Cengwattana office. My visa also has a stamp that says "Please contact the Immigration office for a RE-ENTRY PERMIT Before leaving Thailand".
Guess I will have to go to Chaengwattana office to process the re-entry permit then.
