My husband lives in Singapore and travels to Thailand by air every weekend to visit for 2-3 days . He has been using visa exempt method....but used so many times now getting nervous about potentially being refused entry.
Question is, if he has a METV does he have to have the multiple re entry permit and how would the validity of the visa work if he is flying in and out of Thailand every week?
Any suggestions greatly appreciated.

I think he shouldn't bother about this to much. Wait till immigration question him. Then he can tell them he visit his wife and show them his Singapore wp.

I think, seeing the time spent in Thailand, the best option is the Multiple Entry Tourist Visa, so he can come and go as much as he likes during the validity of 6 months of this visa.

Your husband can get a non o spouse visa. If your company will do it for him. That way he can go in and out easier.
