No, the difference:
With the Single Entry Non Immigrant B visa you can stay until the last day of your Admitted to Stay date stamped in your passport
With the Multiple Entry Non Immigrant B visa you can keep on using it until the valid until date mentioned on the visa and on each entry before that date you get 90 days of stay.
Only the stamp Extension of Stay based on working, which you get at a local Immigration Office just before your Admitted to Stay period ends, needs to be cancelled if you stop working.
